- How it works
- Tenofovir disoproxil fumarate is an oral prodrug hydrolyzed to tenofovir and then phosphorylated intracellularly to the active metabolite tenofovir diphosphate, an acyclic nucleotide analog of adenosine 5'-monophosphate. It competes with the natural substrate for incorporation into viral DNA by reverse transcriptase; because it lacks a 3'-hydroxyl group, its incorporation terminates the growing DNA chain, halting viral replication. This mechanism underlies its activity against both HIV-1 reverse transcriptase and hepatitis B virus polymerase.
- Therapeutic applications
- Indicated in combination with other antiretroviral agents for treatment of HIV-1 infection in adults and pediatric patients 2 years of age and older weighing at least 10 kg. Also indicated for treatment of chronic hepatitis B virus infection in adults and pediatric patients 2 years of age and older weighing at least 10 kg.
