- A lab-made antibody that latches onto the EGFR growth receptor on cancer cells, physically blocking the growth signals that would otherwise tell the tumor to keep dividing.
- How it works
- Panitumumab is a fully human IgG2 monoclonal antibody directed against EGFR. It binds EGFR on both normal and tumor cells with high affinity, competitively blocking endogenous ligand binding and preventing receptor autophosphorylation and downstream activation of proliferative signaling pathways. Because the effect depends on intact EGFR-to-RAS signaling, tumors with activating RAS mutations bypass this blockade, restricting use to RAS wild-type tumors.
- Therapeutic applications
- Indicated for metastatic colorectal cancer with wild-type RAS tumors, as first-line therapy in combination with chemotherapy or as monotherapy after progression on prior chemotherapy. Also indicated in combination with sotorasib for KRAS G12C-mutated disease after prior chemotherapy.
