- How it works
- Brentuximab vedotin is an antibody-drug conjugate composed of a chimeric IgG1 monoclonal antibody directed against CD30, linked via a protease-cleavable linker to the microtubule-disrupting agent monomethyl auristatin E (MMAE). The antibody binds CD30 expressed on lymphoma cells, and the complex is internalized by receptor-mediated endocytosis. Following trafficking to the lysosome, proteolytic cleavage of the linker releases free MMAE intracellularly, which binds tubulin and disrupts the microtubule network, arresting the cell cycle and inducing apoptosis.
- Therapeutic applications
- FDA-indicated for classical Hodgkin lymphoma across multiple treatment settings including newly diagnosed and relapsed/refractory disease, and for systemic anaplastic large cell lymphoma and other CD30-expressing peripheral T-cell and B-cell lymphomas.
