- How it works
- Sarilumab is a recombinant human IgG1 monoclonal antibody that binds with high affinity to both the soluble and membrane-bound forms of the interleukin-6 receptor, competitively inhibiting IL-6-mediated signal transduction. IL-6 is a pro-inflammatory cytokine implicated in synovial inflammation and cartilage degradation. By blocking receptor engagement, sarilumab prevents downstream JAK/STAT3 activation, reducing production of acute-phase inflammatory markers within about two weeks.
- Therapeutic applications
- FDA-approved for moderately to severely active rheumatoid arthritis in adults with inadequate response to DMARDs, polymyalgia rheumatica in adults with inadequate response to corticosteroids, and active polyarticular juvenile idiopathic arthritis in pediatric patients weighing 63 kg or more.
