Golimumab
Simponi
- Mechanism of action
- TNF-alpha inhibitor (mAb)
- In plain language
- A lab-made antibody that neutralizes TNF-alpha, an inflammation-driving protein, to calm autoimmune conditions.
- How it works
- Golimumab is a fully human IgG1 monoclonal antibody that binds with high affinity to both soluble and transmembrane forms of TNF-alpha, preventing the cytokine from engaging its cell-surface receptors. This neutralization blocks downstream TNF-mediated signaling, including expression of adhesion molecules that recruit leukocytes into inflamed tissue and secretion of proinflammatory cytokines. By dampening this inflammatory cascade, golimumab reduces synovial and articular inflammation in rheumatic diseases.
- Therapeutic applications
- FDA-approved for rheumatoid arthritis in combination with methotrexate, psoriatic arthritis alone or with methotrexate, ankylosing spondylitis, and moderately to severely active ulcerative colitis in adults and pediatric patients weighing at least 15 kg.
- Class
- Monoclonal antibody
- Therapeutic area
