How it works
Belimumab is a human IgG1 monoclonal antibody that binds soluble B-lymphocyte stimulator (BLyS, also known as BAFF), preventing it from engaging its on B cells. By neutralizing this survival cytokine, belimumab reduces the survival of B cells, including autoreactive clones, and decreases their differentiation into immunoglobulin-secreting plasma cells. It does not directly bind B cells or deplete them via cytotoxicity, distinguishing its mechanism from B-cell-depleting agents like rituximab.
Therapeutic applications
Indicated in patients aged 5 years and older for active systemic lupus erythematosus as add-on therapy to standard treatment, and for active lupus nephritis, also as add-on therapy.
