- A lab-made antibody that blocks IL-17RA, a docking site on cells that several inflammation-driving proteins need to trigger the overactive skin-cell turnover seen in psoriasis.
- How it works
- Brodalumab is a human IgG2 monoclonal antibody that selectively binds interleukin-17 receptor A (IL-17RA), a subunit shared by several IL-17 receptor complexes. By occupying IL-17RA, it blocks downstream signaling from IL-17A, IL-17F, IL-17C, the IL-17A/F heterodimer, and IL-25. This differs from ligand-targeting IL-17 inhibitors like secukinumab, which block the individual signal proteins rather than the shared receptor, giving brodalumab a broader cytokine-blocking footprint.
- Therapeutic applications
- Indicated for the treatment of moderate-to-severe plaque psoriasis in adults who are candidates for systemic therapy or phototherapy and who have failed to respond, or have lost response, to other systemic therapies. Carries a boxed warning for suicidal ideation and behavior and is only available through a restricted REMS program.
