- A lab-made antibody that latches onto CD38, a marker found in high amounts on multiple myeloma cells, flagging those cells for destruction by the immune system.
- How it works
- Daratumumab is a human IgG1 kappa monoclonal antibody that binds CD38, a transmembrane glycoprotein highly expressed on multiple myeloma cell surfaces. Binding triggers direct tumor cell apoptosis via Fc-mediated cross-linking. It also recruits immune-mediated killing mechanisms, including complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity, to eliminate CD38-expressing malignant plasma cells.
- Therapeutic applications
- FDA-approved for adult multiple myeloma across multiple settings, including combination therapy for newly diagnosed transplant-eligible and transplant-ineligible patients, combination therapy for relapsed/refractory disease, and as monotherapy in heavily pretreated patients.
